Flow Cytometric Analysis of DNA Content and c-myc Oncoprotein Expression in Uterine Cervical Cancer


Abstract
To evaluate the possible role of abnormal DNA content and c-myc oncoprotein overexpression as a prognostic indicator, 65 uterine cervical squamous cell carcinomas and 8 nonneoplastic cervices were retrived from the surgical materials. For DNA
analysis,
the nuclear suspension was stained with propidium iodide and analyzed by flow cytometry. Detection of c-myc oncoprotein was done by fluorimetrically using flow cytometry and immunohistochemically by labeled streptavidin(LSAB) method. DNA plidy,
cell
proliferative activity and c-myc oncoprotein expression were evaluated with histologic malignancy grade, clinical stage. stage. status of lymph node metastasis and recurrence.
@ES The obtained results are summarized as follows;
@EN 1. DNA aneuploidy was found in 26 cases of 65(40%) cervical cancers. There was no correlation between DNA ploidy and clinical stage, however, G2M fraction and proliferative index were significantly higher in invasive carcinoma than in
microinvasive
carcinoma.
2. The proportions of the c-myc positive cells were higher in cervical carcinomas than in nonneoplastic cervices, however, there was no difference between clinical stages. The intensity of oncoprotein immunostaining was higher in invasive
carcinomas
than in microinvasive carcinomas.
3. There was no correlation of histologic malignancy grade with DNA ploidy and proliferative activity, however, the tumors in higher grade malignancy showed more intense immunostaining and positive nuclear area for c-myc oncoprotein than in lower
grade.
The histologic malignancy grade of tumor was positively correlated with clinical stages.
4. DNA aneuploidy and high proliferative activity were related with neither lymph node metastasis nor recurrence. Only the intensity and positive nuclear area for c-myc oncoprotein immunostaining were higher in the cases of lymph node metastasis.
